Streszczenie
Swoista odpowiedź immunologiczna wywiera wpływ na przebieg chorób nowotworowych, między innymi za pośrednictwem szlaku zaprogramowanej śmierci komórki z udziałem limfocytów T posiadających receptor PD-1. Komórki nowotworowe posiadające na swojej powierzchni ligand PD-L-1 przyłączają się do receptora PD-1 na limfocytach T i hamują ich funkcje obronne. Przeciwciała monoklonalne, blokując PD-1, zapobiegają łączeniu się ligandu z receptorem, co przywraca aktywność przeciwnowotworową limfocytów T.
Niwolumab i pembrolizumab są przeciwciałami monoklonalnymi anty-PD-1, zarejestrowanymi do leczenia wielu rodzajów nowotworów, w tym nawrotowego chłoniaka Hodgkina. Przeciwciała anty-PD-1 są również skuteczne w innych rodzajach chłoniaków, co potwierdzają trwające badania kliniczne.
Przeciwciała anty-PD-1 wykazują akceptowalny profil bezpieczeństwa. Rzadkie działania niepożądane zwykle wiążą się z nadmierną aktywacją układu odpornościowego.
