The aging of hematopoietic stem cells and white blood cells

Abstract

The hematopoietic system does not escape the negative effects of aging. Aging of the hematopoietic cells (HSC) compartment causes marked shift in representation of three main subsets (Lymphoid-biased, Myeloid-biased and Balansed-biased) and myeloid biased cells accumulate in the aged bone marrow. Aged HSCs show the self-reneval not complete and the reduction of activity. In the aging of the hematopoiesis both stem cells intrinsic and extrinsic (microenvironmental) factors are involved. Simultaneously the remodeling of the innate immune system is evidenced and defective activation, decreased chemotaxis as well as phagocytosis and intracellular killing of pathogenes by neutrophils and macrophages have been described. Also NK cells show decreased cytotoxicity and dendritic cells — weak antigen presenting process. Immunosenescence of cellular and humoral adaptive response manifested in the reduction of T and B precursors and mature lymphocytes number, especially T CD4+ naïve cells. This remodeling of immune system is connected with impaired reaction for vaccination and increased susceptibility to infections. In aging the concentration of pro-infl ammatory cytokins in peripheral blood is increased.
