The range of clinical effectiveness of clozapine treatment

Abstract

An analysis of clozapine effectiveness in the treatment of various psychiatric disorders was presented. Clozapine is an archetype of the whole class of so-called atypical antipsychotics and has become the reference compound for their clinical effectiveness. The main indications for the clozapine treatment ranges from schizophrenia and schizoaffective disorder, especially suicidal and refractory forms, to recently developing indications to treat affective and psychoorganic syndromes. Studies show consistent findings in favour of clozapine in negative and disorganized dimensions of schizophrenia. The drug is effective and well-tolerated both in young and elderly patients. All haematological, cardiovascular, convulsive and metabolic side-effects should be strictly monitored during clozapine treatment. However there are also benefits of clozapine tolerability, like minimal or null risk of EPS and hyperprolactinemia, no evidence of harm to the foetus, and after recovery from NMS, the potential reintroduction of clozapine instead of former antipsychotic.
